On November 13, 2023, Professor Jinpeng Sun's team from Shandong University, in collaboration with Zhenhua Shao's team from Sichuan University, Gan Li's team from Shanghai Jiao Tong University School of Medicine, and Yue Wang's team from Shandong First Medical University, published an online research paper titled "Structural and signaling mechanisms of TAAR1 enabling preferential agonist design" in the journal Cell. The research team conducted a comprehensive analysis of the characteristics of various endogenous amines and SEP363856 to activate multiple G protein signals of TAAR1. They investigated the molecular mechanism and pharmacological properties associated with the activation of the TAAR1-Gs/Gq pathway. Furthermore, they successfully developed a small molecule agonist called ZH8651 that activates both Gs and Gq pathways simultaneously. The efficacy of this compound was verified in improving schizophrenia symptoms using a mouse model. This study provides valuable insights for designing candidate compounds and developing targeted treatments for schizophrenia.
