On April 10, Professor Sun Jinpeng’s team from Shandong University, Professor Alatan Gaole’s team from Inner Mongolia University, Professor Yu Xiao’s team from Shandong University, and Professor Xia Ming’s team from Shandong First Medical University jointly published an online research paper titled “Structural basis and biased signaling of proton sensation by GPCRs mediated by extracellular histidine rearrangement” in Molecular Cell (with a concurrent special commentary published in Molecular Cell). By comprehensively applying multiple approaches, including pharmacology, computational biology, and biochemical experimental methods, this study systematically deciphered the molecular mechanisms underlying proton sensing, signal transduction, and downstream preferential signaling pathway activation of two human proton-sensing GPCRs (GPR4 and GPR68), providing an important theoretical basis for the development of clinical drugs targeting proton-sensing GPCRs.
