Research programme:the roles and mechanism of CUL4B in regulating adipocyte differentiation
Research Interest:
Obesity has been becoming a disease very harmful to our health in modern world. Among the many causes, abnormal adipocyte differentiation is a significant factor.
Previous studies have shown that mutations in CUL4B, which encodes a ubiquitin E3 ligase subunit, cause an X-linked mental retardation syndrome associated with central obesity. However, the roles of CUL4B in obesity and lipid metabolism are totally unknown. Our studies showed that Cul4b deletion dramatically enhance adipogenic differentiation of mesenchymal stem cells. We believe that the function of CUL4B in adipocyte differentiation could explain the phenotype that CUL4B mutation causes obesity. To test this hypothesis, we will use Cul4b knock out mouse and in vitro adipocyte differentiation assay to investigate the roles and molecular mechanism of CUL4B in regulating adipocyte differentiation. This study will help us understand the incidence of obesity and provide new targets for obesity treatment.
